Real Time RT-PCR02:57

Real Time RT-PCR

61.7K
Real-time reverse transcription-polymerase chain reaction, or Real-time RT-PCR, is an analytical tool used to determine the expression level of target genes. The method involves converting mRNA to complementary DNA with the help of an enzyme known as reverse transcriptase, followed by the PCR amplification of the cDNA. These two processes can be performed simultaneously in a single tube or separately as a two-step reaction.
The real-time quantification of the number of amplified products is...

Area of Science:

  • Molecular Biology
  • Infectious Diseases
  • Medical Diagnostics

Background:

  • The COVID-19 pandemic highlighted the need for rapid and efficient diagnostic tools.
  • Existing SARS-CoV-2 detection methods, like RT-PCR, often involve lengthy sample preparation steps.
  • The demand for nucleic acid-based testing strains reagent availability and laboratory capacity.

Purpose of the Study:

  • To evaluate the performance of a novel diagnostic test, DirectDetect SARS-CoV-2 Direct Real-time RT-PCR.
  • To assess the test's ability to simplify and accelerate SARS-CoV-2 detection.
  • To determine the feasibility of DirectDetect for various sample types, reducing time and reagent requirements.

Main Methods:

  • A preliminary study was conducted on 24 respiratory samples from 14 SARS-CoV-2-positive patients.
  • The DirectDetect SARS-CoV-2 Direct Real-time RT-PCR test was evaluated.
  • Performance was compared against standard clinical laboratory tests.

Main Results:

  • DirectDetect eliminates the need for viral transport medium and RNA extraction.
  • Sample preparation time is reduced from hours to minutes.
  • Positive agreement rates were high: 94.7% for dry swabs, 100% for treated samples, and 94.7% for untreated samples compared to clinical lab tests.

Conclusions:

  • DirectDetect SARS-CoV-2 Direct Real-time RT-PCR is a promising diagnostic tool for COVID-19.
  • The test simplifies the diagnostic workflow, significantly reducing time and reagent usage.
  • Further validation with larger sample sizes is recommended to confirm its clinical utility.
