Summary

Nitrite exposure can cause infertility in female mice, particularly with higher doses and repeated exposure. This infertility is linked to increased ovarian cell apoptosis and oxidative stress.

Background:

Purpose of the Study:

  • To investigate the effects of nitrite exposure on female mouse fertility.
  • To elucidate the underlying mechanisms of nitrite-induced infertility, focusing on apoptosis and oxidative stress.

Main Methods:

  • Adult female C57BL/6J mice were exposed to varying nitrite doses and durations.
  • Ovarian tissues were analyzed using hematoxylin and eosin staining, TUNEL assay, immunofluorescence, and western blotting.

Main Results:

  • Nitrite exposure significantly increased infertility rates in a time-dependent manner.
  • Increased atretic follicles, granulosa cell apoptosis (TUNEL-positive cells), and overexpression of caspase 8, c-Fos, and inducible nitric oxide synthase (iNOS) were observed.

  • Nitrite exposure induces infertility in mice.
  • Apoptosis and oxidative stress are key mechanisms mediating nitrite-induced infertility.
