Summary

Mast cells in lung cancer (NSCLC) show distinct types within tumors. While one type doesn't improve prognosis, higher total mast cell numbers correlate with better survival for cancer patients.

Background:

  • Mast cells (MCs) are innate immune cells in the tumor microenvironment (TME).
  • Their role in cancer and prognostic value are poorly understood, with conflicting associations.
  • Few studies have characterized MCs within the tumor immune infiltrate.

Purpose of the Study:

  • To characterize mast cell populations in the tumor microenvironment of Non-Small Cell Lung Cancer (NSCLC).
  • To investigate the phenotype and potential prognostic value of different mast cell subsets in NSCLC.

Main Methods:

  • Multicolor flow cytometry and confocal microscopy were used.
  • Analysis was performed on a surgical cohort of 48 NSCLC patients.
  • Mast cells were compared between tumor tissue and matching non-lesional lung areas.

Main Results:

  • Tumor-associated mast cells (TAMCs) exhibit a distinct phenotype compared to non-lesional MCs.
  • Two TAMC subsets were identified based on CD103 expression: CD103+ and CD103-.
  • CD103+ TAMCs showed signs of maturation, T cell interaction potential, and proximity to cancer cells.
  • High frequency of total TAMCs correlated with improved overall and progression-free survival.
  • No prognostic advantage was observed for the CD103+ TAMC subset specifically.

Conclusions:

  • Tumor-associated mast cells in NSCLC are heterogeneous, with distinct subsets.
  • Mast cell subsets may have different roles and prognostic implications in NSCLC.
  • Further research into MC subsets is crucial for patient stratification and management in lung cancer.
