Researchers developed a novel molecular imprinting technique to create nanoparticles that activate acetylcholinesterase (AChE) enzyme activity. These nanoMIPs show promise for treating enzyme deficiencies and in medical emergencies.
Area of Science:
- Biochemistry
- Nanotechnology
- Enzyme kinetics
Background:
- Enzyme activity modulation is crucial for biological pathways.
- Pharmaceutical strategies focus on inhibitors, but activators are less developed.
- Acetylcholinesterase (AChE) is a key enzyme with implications in various physiological and pathological processes.
Purpose of the Study:
- To develop novel methods for promoting enzyme activation.
- To identify surface epitopes on AChE for targeted activation.
- To create molecularly imprinted nanoparticles (nanoMIPs) for enzyme modulation.
Main Methods:
- Innovative epitope mapping using molecular imprinting.
- Synthesis of nanoMIPs targeting identified AChE surface epitopes.
- Enzyme activity assays to quantify nanoMIP effects on native and inhibited AChE.
Main Results:
- Four surface epitopes of AChE were successfully identified.
- Synthesized nanoMIPs significantly increased AChE enzymatic activity (up to 47×).
- nanoMIPs restored inhibited AChE activity from 11% to 73%.
Conclusions:
- Molecular imprinting provides a new strategy for enzyme activation.
- nanoMIPs demonstrate allosteric modulation by stabilizing protein conformation.
- This approach offers potential therapeutic applications for enzyme deficiency diseases and poisoning.

Reversible inhibitors like edrophonium bind to a specific part of the enzyme called the anionic catalytic site. They form noncovalent bonds, which means they are not strongly attached to the enzyme. This creates a temporary and less stable enzyme–inhibitor complex,...
