Melatonin, a hormone, protects against lung damage caused by excess copper exposure. This study shows melatonin reduces oxidative stress and inflammation in copper-exposed rats, suggesting a therapeutic role.
Area of Science:
- Toxicology
- Pharmacology
- Biochemistry
Background:
- Copper (Cu) is essential but excess intake causes oxidative stress and inflammation.
- Melatonin, the hormone of darkness, has known antioxidant and anti-inflammatory properties.
- The protective effects of melatonin against copper-induced lung injury are not well understood.
Purpose of the Study:
- To investigate the potential protective effects of melatonin against copper-induced lung damage.
- To assess melatonin's efficacy in mitigating copper-induced oxidative stress and inflammation in rat lungs.
Main Methods:
- Female Sprague Dawley rats were exposed to 250 ppm copper in drinking water for 16 weeks.
- Rats received melatonin (5 and 10 mg/kg) via intraperitoneal injection from week 13 to 16.
- Lung tissue damage was evaluated using oxidative stress markers, metal analysis, and histological examination.
Main Results:
- Copper exposure significantly increased oxidative stress markers and copper accumulation in lung tissue.
- Histological analysis revealed severe tissue injury and inflammatory cell infiltration in copper-exposed rats.
- Melatonin treatment attenuated oxidative stress, reduced inflammation, and decreased collagen deposition compared to copper-exposed rats.
Conclusions:
- Melatonin demonstrates significant antioxidant and anti-inflammatory effects in the context of copper-induced lung injury.
- Melatonin may prevent lung damage by inhibiting reactive oxygen species (ROS)-mediated oxidative stress and inflammation.
- These findings suggest melatonin as a potential therapeutic agent for copper-related pulmonary conditions.
