Summary
Adult mice generate two main types of gamma delta (γδ) T cells: immature cells that mature post-thymic export and a smaller group that matures within the thymus. Resident thymic γδ T cells were also identified.
Area of Science:
- Immunology
- T cell biology
- Developmental immunology
Background:
- Gamma delta (γδ) T cells have distinct developmental pathways in fetal vs. adult thymus.
- The coordination of γδ T cell differentiation and effector function in adult thymus is not well understood.
Purpose of the Study:
- To investigate the relationship between maturation and thymic export of adult γδ T cells in mice.
- To determine if γδ T cell maturation and effector function acquisition occur within the adult thymus or peripherally.
Main Methods:
- Utilized the Rag2pGFP mouse model to track γδ T cell maturation.
- Analyzed γδ T cell populations in thymus and periphery using flow cytometry (CD24, GFP markers).
- Performed adoptive transfer experiments to assess in vivo maturation potential.
Main Results:
- Immature (CD24+) γδ thymocytes highly expressed GFP, while mature (CD24-) cells had low expression.
- Most peripheral γδ T cells and recent thymic emigrants (RTEs) were immature (CD24+, GFP+).
- Adoptive transfer confirmed immature γδ thymocytes mature outside the thymus; mature cells rarely recirculated.
Conclusions:
- Adult mice export two main γδ T cell subsets: immature cells maturing peripherally and a minor subset maturing in the thymus.
- A population of mature, resident γδ T cells persists in the adult thymus.
- These findings clarify the generation and export dynamics of the adult γδ T cell compartment.
