    Summary

    This study reveals a significant shift in immunoglobulin A (IgA) subclass distribution within the human intestine. IgA1-producing cells are more common in the jejunum, while IgA2-producing cells dominate the lower intestine.

    Background:

    • Immunoglobulin A (IgA) is crucial for mucosal immunity.
    • IgA exists in two subclasses, IgA1 and IgA2, with distinct structural and functional properties.
    • Understanding IgA subclass distribution in the human intestine is vital for comprehending mucosal immune responses.

    Purpose of the Study:

    • To investigate the differential distribution of IgA1- and IgA2-containing cells in three distinct regions of the human intestine: upper jejunum, terminal ileum, and large bowel.
    • To determine if there is a regional variation in the relative abundance of IgA1 and IgA2 subclasses throughout the intestine.

    Main Methods:

    • Analysis of 27 normal intestinal tissue specimens from the upper jejunum, terminal ileum, and large bowel.
    • Immunoperoxidase staining using specific monoclonal antibodies to identify IgA1- and IgA2-containing cells.
    • Quantitative assessment of IgA1+ and IgA2+ cell counts and calculation of the IgA1+/IgA2+ ratio in each intestinal segment.

    • IgA1-producing cells predominate in the upper jejunum.
    • IgA2-producing cells are more prevalent in the intestine distal to the terminal ileum, including the large bowel.
