Aminoglutethimide in advanced prostatic carcinoma.

    Summary

    Aminoglutethimide offers a treatment option for advanced prostate cancer resistant to standard hormonal therapies. This study found that suppressed gonadotrophin levels correlated with patient benefit, despite some treatment toxicity.

    Background:

    • Advanced prostate cancer often becomes resistant to orchiectomy and oestrogen therapy.
    • Identifying effective treatments for hormone-refractory prostate cancer is crucial.

    Purpose of the Study:

    • To evaluate the efficacy and toxicity of aminoglutethimide in patients with advanced prostate cancer resistant to primary hormonal manipulation.
    • To explore potential biomarkers correlating with treatment response.

    Main Methods:

    • A cohort of 34 patients with advanced prostate cancer received aminoglutethimide treatment.
    • Patient outcomes were assessed based on pain, performance status, and National Prostate Cancer Project (NPCP) criteria.
    • Hormone levels (gonadotrophins, testosterone, dehydroepiandrosterone, oestradiol, prolactin) were monitored.
