Inflammatory signals enhance mesenchymal stem cell (MSC) extracellular vesicles (EVs) for tissue repair. TNFα-preconditioned MSC EVs improved bone regeneration by reducing inflammation and modulating macrophage responses, suggesting a role for EV miRNAs.
Area of Science:
- Cell Biology
- Immunology
- Regenerative Medicine
Background:
- Mesenchymal stem cells (MSCs) possess immunomodulatory properties crucial for tissue repair.
- Extracellular vesicles (EVs) secreted by MSCs mediate many of these paracrine effects.
- The impact of inflammatory environments on MSC EV function in tissue repair remains incompletely understood.
Purpose of the Study:
- To investigate the effect of the inflammatory cytokine TNFα on MSCs and their secreted EVs.
- To evaluate the immunomodulatory capacity of TNFα-preconditioned MSC EVs in vitro and in vivo.
- To explore the potential mechanisms, including miRNA changes, underlying enhanced MSC EV function.
Main Methods:
- MSCs were preconditioned with TNFα.
- EVs were isolated and characterized.
- In vitro studies involved co-culture with polarized (M1/M2) macrophages.
- In vivo studies utilized a rat calvarial defect model.
- EV miRNA content was analyzed.
Main Results:
- TNFα preconditioning did not alter MSC EV physical characteristics but enhanced their immunomodulatory properties.
- Preconditioned MSC EVs suppressed M1 macrophage markers and promoted M2 markers in vitro.
- In vivo, TNFα-preconditioned MSC EVs reduced inflammation and enhanced bone formation in a calvarial defect model.
- Significant changes in anti-inflammatory miRNAs were observed in preconditioned EVs.
Conclusions:
- Exposure to inflammatory signals like TNFα enhances the immunomodulatory function of MSC EVs for tissue repair.
- TNFα-preconditioned MSC EVs show promise for improving bone regeneration by modulating inflammatory responses.
- EV-derived miRNAs may play a key role in the enhanced immunomodulatory activity of preconditioned MSC EVs.
