Area of Science:

  • Neuroscience
  • Pharmacology
  • Cell Biology

Background:

  • Depression incidence is increasing globally.
  • Developing effective treatments for depression is a priority.
  • Investigating natural compounds for neuroprotective effects is crucial.

Purpose of the Study:

  • To evaluate the protective effects of Schisandrin on a corticosterone (CORT)-induced PC12 cell model of depression.
  • To elucidate the underlying mechanisms of Schisandrin's action in this cellular model.

Main Methods:

  • PC12 cells were treated with CORT to induce a depressive phenotype.
  • Cell viability was assessed using the MTT assay.
  • Lactate dehydrogenase (LDH) activity, apoptosis rates, and mitochondrial membrane potential were measured.
  • Western blotting was used to analyze the expression of apoptosis-related proteins (Caspase-3, Bax, Bcl-2).

Main Results:

  • Schisandrin significantly increased cell viability and reduced LDH content in CORT-treated PC12 cells.
  • Schisandrin treatment decreased mitochondrial membrane potential.
  • The compound effectively reduced apoptosis by inhibiting pro-apoptotic proteins (cleaved Caspase-3, Bax) and increasing anti-apoptotic protein (Bcl-2).

Conclusions:

  • Schisandrin exhibits significant protective effects against CORT-induced depression in PC12 cells.
  • These protective effects are mediated by the inhibition of cellular apoptosis.
  • Schisandrin demonstrates potential as a therapeutic agent for depression.
