Area of Science:

  • Molecular Biology
  • Genetics
  • Virology

Background:

  • SmartAmp-Eprimer Binary code (SEB) Genotyping is a novel isothermal amplification technique.
  • It is designed for rapid genotyping of variable targets.

Purpose of the Study:

  • To present the proof of concept for SEB Genotyping.
  • To demonstrate its application in Hepatitis B Virus (HBV) genotyping.

Main Methods:

  • In silico sequence alignment and computational analysis identified key target regions.
  • SmartAmp primer sets were designed to generate On/Off fluorescence signals, creating a binary code for each genotype.

Main Results:

  • Applied to HBV, 4 targets were selected to generate a unique binary code for each of the 8 main genotypes (A-H).
  • The method demonstrated accurate results quickly.

Conclusions:

  • SEB Genotyping is a new method for complex, highly variable targets.
  • It can be adapted for genotyping other pathogens or diseases with multiple mutations.
  • The technique enables rapid, accurate results for informed treatment selection.
