Area of Science:

  • Mitochondrial Biology
  • Insect Physiology
  • Biochemistry

Background:

  • Mitochondria isolation from insects is challenging due to low purity and activity.
  • Existing methods lack comprehensive validation across insect orders.

Purpose of the Study:

  • To develop and validate a robust method for isolating highly pure and active insect mitochondria.
  • To assess the integrity and activity of isolated mitochondria under various storage conditions.

Main Methods:

  • Combined differential centrifugation and discontinuous Nycodenz density gradient centrifugation.
  • Validated across Coleoptera, Hymenoptera, and Blattaria insect orders.
  • Purity assessed by Western blot and TEM; activity/integrity by TEM, staining, fluorescence, and flow cytometry.

Main Results:

  • Achieved high purity by removing non-mitochondrial contaminants (nucleus, cytosol, ER).
  • Demonstrated no loss of mitochondrial integrity or activity post-isolation.
  • Mitochondrial activity is preserved at -20°C, but compromised at 4°C and 25°C.

Conclusions:

  • The developed method yields highly pure and active insect mitochondria.
  • The protocol is widely applicable across diverse insect species.
  • Optimal storage at -20°C is recommended to maintain mitochondrial function for experiments.
