Angiotensin II (Ang II) impairs insulin signaling in fat cells by inhibiting insulin receptor phosphorylation and glucose uptake. This effect is mediated by protein kinase C (PKC) activation, suggesting a link to insulin resistance.
Area of Science:
- Endocrinology
- Molecular Biology
- Metabolic Research
Background:
- Angiotensin II (Ang II) regulates insulin signaling in cardiovascular and metabolic tissues.
- The specific role of Ang II in adipose cells' insulin signaling is not fully understood.
Purpose of the Study:
- To investigate the effect of Ang II on insulin signaling pathways in 3T3-L1 adipocytes.
- To elucidate the mechanism by which Ang II influences insulin-stimulated glucose uptake and insulin receptor (IR) activity.
Main Methods:
- Utilized 3T3-L1 adipocytes and isolated adipocytes for experiments.
- Examined insulin-induced insulin receptor (IR) and insulin receptor substrate-1 (IRS-1) phosphorylation (tyrosine and serine).
- Assessed Akt activation, glucose uptake, and protein kinase C (PKC) isoform activity and association with the IR.
Main Results:
- Ang II inhibited insulin-stimulated tyrosine phosphorylation of IR and IRS-1, Akt activation, and glucose uptake in a time-dependent manner.
- Ang II increased serine phosphorylation of the IR, but not IRS-1, suggesting IR as the primary site of desensitization.
- PKC inhibition restored insulin signaling, and Ang II activated multiple PKC isoforms (PKCα/βI/βII/δ), with PKCβII showing high interaction with the IR.
Conclusions:
- Angiotensin II negatively impacts insulin signaling in adipocytes, potentially leading to insulin resistance.
- Protein kinase C (PKC) activation by Ang II is a key mechanism underlying this insulin desensitization in fat cells.
