Oral Hypoglycemic Agents: α-Glucosidase Inhibitors

253
α-glucosidase inhibitors, including acarbose (Precose), miglitol (Glyset), and voglibose (Voglib) (primarily available in Asia), are drugs that control blood sugar levels by delaying the digestion of starch and disaccharides. They achieve this by inhibiting α-glucosidase enzymes in the intestine, which slow the absorption of carbohydrates in the intestine, which in turn leads to a prolonged release of the glucoregulatory hormone GLP-1 from intestinal L-cells.
Acarbose and miglitol are...

Area of Science:

  • Phytochemistry
  • Natural Product Chemistry

Background:

  • Agrimonia pilosa is a plant with potential medicinal properties.
  • Understanding its chemical constituents is crucial for exploring its biological activities.

Purpose of the Study:

  • To isolate and identify lignan compounds from the 60% ethanol extract of Agrimonia pilosa.
  • To characterize a novel lignan and report the first isolation of thirteen lignans from this plant species.

Main Methods:

  • Extraction of Agrimonia pilosa using 60% ethanol.
  • Isolation of compounds via column chromatography (silica gel, ODS, MCI) and preparative high-performance liquid chromatography (HPLC).
  • Structure elucidation using physicochemical properties and spectral data analysis.

Main Results:

  • Thirteen lignans were successfully isolated and identified.
  • A novel lignan (compound 1) was discovered.
  • Compounds 1-13 represent the first reported isolation of these lignans from the Agrimonia plant.

Conclusions:

  • The chemical profile of Agrimonia pilosa has been significantly enriched with the identification of thirteen lignans.
  • This study provides a foundation for further research into the biological activities and pharmacodynamic substances of Agrimonia pilosa.
