Ribosome Profiling02:24

Ribosome Profiling

3.6K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

Area of Science:

  • Plant Science
  • Genomics
  • Biochemistry

Background:

  • Rooibos (Aspalathus linearis), a South African endemic, possesses health-promoting phenolic compounds.
  • Distinct wild ecotypes exhibit variations in morphology and biochemistry, with only one cultivated commercially.

Purpose of the Study:

  • To establish non-invasive transcriptome research methodologies for wild rooibos.
  • To generate annotated transcriptomes for diverse rooibos ecotypes.
  • To support future rooibos research and breeding initiatives.

Main Methods:

  • Developed protocols for harvesting, transport, and RNA extraction from challenging plant material.
  • Implemented high-throughput biochemical screening and biocomputational analysis of Illumina sequencing data.
  • Evaluated transcriptome assembly, protein prediction, and annotation pipelines.

Main Results:

  • Established a biochemically characterized sample pool from 44 rooibos ecotypes.
  • Generated four in-depth annotated transcriptomes (approx. 86,000 transcripts each).
  • Successfully extracted high-quality RNA from polyphenol-rich plant tissues.

Conclusions:

  • The developed methodologies enable non-invasive transcriptome research in wild plant species.
  • The generated transcriptomes offer valuable genomic resources for rooibos.
  • This work facilitates future investigations into rooibos's health benefits and breeding potential.
