    Background:

    • Spermatocytic seminoma presents unique clinical and histological features compared to other testicular germ cell tumors.
    • Classical seminomas and teratomas are thought to arise from intratubular carcinoma-in-situ (CIS), often found adjacent to these tumors.
    • DNA analysis of classical germ cell tumors and adjacent CIS reveals aneuploid DNA distribution.

    Purpose of the Study:

    • To investigate the origin of spermatocytic seminoma.
    • To determine the presence of CIS adjacent to spermatocytic seminomas.
    • To analyze the DNA content of spermatocytic tumor cells.

    Main Methods:

    • Histological examination of testicular tissue adjacent to fifteen spermatocytic seminomas.
    • Assessment for the presence of carcinoma-in-situ (CIS).
    • Densitometric DNA content analysis of spermatocytic tumor cells.

    Main Results:

    • Carcinoma-in-situ (CIS) was not observed in any of the twelve specimens with adjacent seminiferous tubules.
    • Nuclear DNA measurements were performed on eleven spermatocytic seminomas.
    • DNA distribution was diploid in five cases, tetraploid in three, and aneuploid in three cases.

    Conclusions:

    • Spermatocytic seminoma lacks adjacent carcinoma-in-situ (CIS).
    • The DNA content of spermatocytic seminoma is frequently diploid or tetraploid, unlike the aneuploid classical germ cell tumors.
    • These findings suggest spermatocytic seminoma originates from a different cell type than other testicular germ cell tumors.
