Summary

Villin protein is a useful marker for gastrointestinal and pancreatobiliary tumors, but its expression in 54 tumor types means it should be used in panels, not alone.

Purpose of the Study:

  • To comprehensively determine villin expression across diverse tumor types.
  • To evaluate villin's utility as an immunohistochemical marker.

Main Methods:

  • Analysis of tissue microarrays with 14,398 tumor samples (118 types) and 608 normal tissue samples.
  • Immunohistochemistry was used to detect villin expression.

Main Results:

  • Villin was detected in 54 of 118 tumor categories, with strong staining in 36.
  • Frequent villin expression observed in colorectal, upper GI, pancreatobiliary, ovarian, yolk sac, and neuroendocrine tumors.
  • Reduced villin expression correlated with advanced pT stage, lymph vessel invasion, and microsatellite instability in colorectal adenocarcinoma.

Conclusions:

  • Villin immunohistochemistry is valuable for identifying tumors of gastrointestinal, pancreatobiliary, and yolk sac origin.
  • Due to expression in 54 tumor categories, villin should be part of a marker panel, not used alone.
