    Summary

This study tracked E. coli enterotoxin production in a large reactor. Active enterotoxin accumulated within six hours, showing potential for industrial-scale enterotoxin preparation.

    Area of Science:

    • Microbiology
    • Biotechnology

    Background:

    • Escherichia coli (E. coli) produces enterotoxins relevant to various applications.
    • Optimizing enterotoxin production is crucial for industrial purposes.

    Purpose of the Study:

    • To investigate the accumulation of E. coli enterotoxin in a large-scale fermentation system.
    • To characterize the properties of the produced enterotoxin and assess its potential for industrial production.

    Main Methods:

    • Cultivation of E. coli strain P-99 in a 30-litre reactor under aerated conditions.
    • Monitoring oxygen utilization, pyruvic acid synthesis and release, and protein accumulation.
    • Characterization of the enterotoxin preparation, including stability, toxin types, and biological activity (mouse limb edema test).

    Main Results:

    • Significant accumulation of active, high-molecular-weight E. coli enterotoxin was observed within a 6-hour cultivation period.
    • The process involved oxygen utilization, pyruvic acid release, and protein synthesis.
    • The resulting enterotoxin preparation demonstrated stability after lyophilization and contained both thermolabile and thermostable toxins, inducing mouse limb edema.

    Conclusions:

    • The study successfully demonstrated the feasibility of accumulating active E. coli enterotoxin in a large-scale reactor.
    • The characterized enterotoxin exhibits properties suitable for industrial production.
    • These findings are significant for the development of industrial processes for active enterotoxin preparations.
