Marine-derived fiscalins show neuroprotective effects against common mechanisms in neurodegenerative diseases (ND). Certain derivatives protect against MPP+ and iron toxicity and modulate P-glycoprotein (P-gp) activity, offering potential for new ND treatments.
Area of Science:
- Marine biotechnology
- Neuroscience
- Pharmacology
Background:
- Neurodegenerative diseases (ND) share common pathogenic mechanisms.
- Marine-derived compounds offer potential therapeutic strategies.
- Fiscalins are marine compounds with potential neuroprotective properties.
Purpose of the Study:
- To evaluate the neuroprotective effects of fiscalin derivatives against MPP+ and iron(III)-induced cytotoxicity in an in vitro model of ND (SH-SY5Y cells).
- To assess the impact of fiscalin derivatives on P-glycoprotein (P-gp) activity, a key efflux pump for neurotoxins.
Main Methods:
- Differentiated SH-SY5Y cells were exposed to MPP+ or iron(III) and 18 fiscalin derivatives at non-cytotoxic concentrations.
- Cytotoxicity was evaluated at 24 and 48 hours post-exposure.
- P-gp transport activity was measured to assess inhibition or activation.
Main Results:
- Fiscalins 1a and 1b demonstrated significant protection against MPP+-induced cytotoxicity.
- Fiscalins 1b, 2b, 4, and 5 showed protective effects against iron(III)-induced cytotoxicity.
- Fiscalins 4 and 5 inhibited P-gp activity, while others (1c, 2a, 2b, 6, 11) modestly increased it.
Conclusions:
- Fiscalin derivatives exhibit promising neuroprotective effects against key neurodegenerative disease stressors.
- These compounds modulate P-gp activity, suggesting potential as P-gp inhibitors or activators.
- Fiscalins represent a valuable source for developing novel therapeutic agents for neurodegenerative diseases.
