Phase Contrast and Differential Interference Contrast Microscopy01:26

Phase Contrast and Differential Interference Contrast Microscopy

8.7K
Phase-Contrast Microscopes
In-phase-contrast microscopes, interference between light directly passing through a cell and light refracted by cellular components is used to create high-contrast, high-resolution images without staining. It is the oldest and simplest type of microscope that creates an image by altering the wavelengths of light rays passing through the specimen. Altered wavelength paths are created using an annular stop in the condenser. The annular stop produces a hollow cone of...

Imaging Biological Samples with Optical Microscopy

5.1K
Optical microscopy uses optic principles to provide detailed images of samples. Antonie van Leeuwenhoek designed the first compound optical microscope in the 17th century to visualize blood cells, bacteria, and yeast cells. In 1830, Joseph Jackson Lister created an essentially modern light microscope. The 20th century saw the development of microscopes with enhanced magnification and resolution.
In optical microscopy, the specimen to be viewed is placed on a glass slide and clipped on the stage...

Super-resolution Fluorescence Microscopy01:37

Super-resolution Fluorescence Microscopy

7.2K
Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been...

Upsampling01:22

Upsampling

302
Managing signal sampling rates is essential in digital signal processing to maintain signal integrity. A decimated signal, characterized by a reduced frequency range due to its lower sampling rate, can be upsampled by inserting zeros between each sample. This upsampling process expands the original spectrum and introduces repeated spectral replicas at intervals dictated by the new Nyquist frequency. To refine this zero-inserted sequence, it is passed through a lowpass filter with a cutoff...

Aliasing

Accurate signal sampling and reconstruction are crucial in various signal-processing applications. A time-domain signal's spectrum can be revealed using its Fourier transform. When this signal is sampled at a specific frequency, it results in multiple scaled replicas of the original spectrum in the frequency domain. The spacing of these replicas is determined by the sampling frequency.
If the sampling frequency is below the Nyquist rate, these replicas overlap, preventing the original...

Magnetic Resonance Imaging01:24

Magnetic Resonance Imaging

6.4K
Magnetic resonance imaging (MRI) is a noninvasive medical imaging technique based on a phenomenon of nuclear physics discovered in the 1930s, in which matter exposed to magnetic fields and radio waves was found to emit radio signals. In 1970, a physician and researcher named Raymond Damadian noticed that malignant (cancerous) tissue gave off different signals than normal body tissue. He applied for a patent for the first MRI scanning device in clinical use by the early 1980s. The early MRI...

    Summary

Combining Aperture Domain Model Image Reconstruction (ADMIRE) with Minimum Variance (MV) beamforming significantly improves ultrasound image quality. This novel approach enhances sizing and contrast ratio, outperforming existing methods in simulations and in vivo data.

    Area of Science:

    • Ultrasound imaging
    • Medical image processing
    • Acoustic signal processing

    Background:

    • Aperture Domain Model Image Reconstruction (ADMIRE) is a frequency-domain beamformer designed to reduce reverberation and clutter.
    • Minimum Variance (MV) beamforming aims to reduce off-axis interference and improve lateral resolution but struggles in high noise and has unevaluated performance with reverberation.
    • Combining ADMIRE and MV beamformers offers a potential synergistic improvement in image quality.

    Purpose of the Study:

    • To evaluate the effectiveness of using ADMIRE as a preprocessing step before MV beamforming.
    • To assess the performance of the combined ADMIRE + MV approach in suppressing noise and improving image quality in simulated and in vivo ultrasound data.
    • To analyze the assumptions of MV beamforming in relation to reverberation clutter.

    Main Methods:

    • Implementation of ADMIRE as a noise suppression pre-processing step.
    • Application of both narrowband and broadband Minimum Variance (MV) beamforming algorithms.
    • Testing on simulated data with reverberation clutter and in vivo curvilinear kidney data.
    • Comparison of ADMIRE + MV against MV alone, ADMIRE alone, and conventional Delay-and-Sum (DAS) beamforming.

    Main Results:

    • ADMIRE + MV demonstrated significant sizing improvements over MV alone (52.1% in simulations, 14.5% in vivo) in reverberation scenarios.
    • The combined approach improved contrast ratio compared to ADMIRE alone (15.1% in simulations, 14.0% in vivo).
    • ADMIRE + MV consistently outperformed DAS, MV, and ADMIRE in both sizing and contrast ratio.

    Conclusions:

    • The ADMIRE + MV combination effectively suppresses noise and enhances image quality in ultrasound.
    • This approach offers substantial improvements in object sizing and contrast ratio, particularly in the presence of reverberation.
    • ADMIRE + MV represents a valuable advancement for ultrasound image reconstruction, outperforming traditional methods.
