Summary
Aberrant microRNA (miRNA) expression, including miR-99b and miR-377, is linked to advanced renal cell carcinoma (RCC). A panel of miRNAs may serve as a prognostic marker for RCC progression.
Area of Science:
- Oncology
- Molecular Biology
- Biomarker Research
Background:
- Renal cell carcinoma (RCC) is a common adult solid tumor with a lack of reliable prognostic biomarkers.
- MicroRNA (miRNA) dysregulation is implicated in RCC progression, but expression patterns remain controversial.
Purpose of the Study:
- To investigate the expression and clinical significance of specific miRNAs (miR-99b, -144, -155, -210, -222, -302a, -377) in RCC.
- To determine if these miRNAs can serve as prognostic markers for RCC.
Main Methods:
- Retrospective analysis of miRNA expression in tumor tissue from 126 clear cell RCC patients.
- Quantitative polymerase chain reaction (qPCR) was used to measure miRNA levels.
Main Results:
- Overexpression of miR-99b, -222, -302a and low miR-155 levels correlated with advanced RCC stage.
- High levels of miR-99b and miR-377 were detected in patients with distant metastases.
- miRNA ratios (miR-99b, -144, -155, -210, -222, -302a) were associated with Fuhrman tumor grade.
Conclusions:
- Aberrant expression of the studied miRNAs is strongly associated with advanced RCC.
- A panel of these miRNAs shows potential as a useful prognostic marker for RCC course.

MicroRNAs
3.1K
MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

lncRNA - Long Non-coding RNAs
8.8K
In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA...
