Master Transcription Regulators02:23

Master Transcription Regulators

7.0K
Master transcription regulators are regulatory proteins that are predominantly responsible for regulating the expression of multiple genes. Often these genes work in concert to drive a  complex process. Activation of a master transcription regulator can lead to a cascade of transcriptional activation necessary for that outcome. These regulators can directly bind to the regulatory sequences of the various genes involved, or they can indirectly regulate transcription by binding to regulatory...

Epigenetic Regulation

31.3K
Epigenetic mechanisms play an essential role in healthy development. Conversely, precisely regulated epigenetic mechanisms are disrupted in diseases like cancer.

Summary

This study identified seven N6-methyladenosine (m6A) methylation regulator genes that link prostate cancer and periodontitis. The FMR1 gene emerged as a key prognostic indicator for prostate adenocarcinoma (PRAD).

Area of Science:

  • Oncology
  • Genetics
  • Epigenetics
  • Oral Biology

Background:

  • Prostate adenocarcinoma (PRAD) and periodontitis (PD) are prevalent diseases with potential shared molecular mechanisms.
  • N6-methyladenosine (m6A) methylation is a crucial epigenetic modification involved in various biological processes, including cancer development.

Purpose of the Study:

  • To identify shared N6-methyladenosine (m6A) methylation regulator genes between prostate adenocarcinoma (PRAD) and periodontitis (PD).
  • To investigate the prognostic value of these shared genes in PRAD.

Main Methods:

  • Differential gene expression analysis of PRAD and PD datasets.
  • Identification of overlapping differentially expressed m6A-related genes (crosstalk genes).
  • Survival analysis, lasso regression, and protein-protein interaction (PPI) network construction.

Main Results:

  • Seven m6A-related genes (ALKBH5, FMR1, IGFBP3, RBM15B, YTHDF1, YTHDF2, ZC3H13) were identified as crosstalk genes.
  • FMR1 was identified as a significant prognostic indicator for PRAD survival.
  • Four genes (ALKBH5, IGFBP3, RBM15B, FMR1) were highlighted by regression and survival analyses for their influence on risk levels.

Conclusions:

  • Seven m6A methylation regulators act as crosstalk genes between PRAD and PD.
  • FMR1 holds prognostic significance for PRAD, suggesting a potential link between m6A regulation, periodontitis, and prostate cancer progression.
