D-carnitine and tetradecylglycidic acid (TDGA) significantly inhibit intestinal absorption of palmitic acid in rats. This suggests a role for carnitine palmitoyltransferase in fatty acid uptake in the intestine.

    Area of Science:

    • Biochemistry
    • Physiology
    • Gastroenterology

    Background:

    • Fatty acid absorption is crucial for energy metabolism.
    • Carnitine palmitoyltransferase (CPT) enzymes are involved in fatty acid transport into mitochondria.
    • The role of CPT in intestinal fatty acid absorption is not fully understood.

    Purpose of the Study:

    • To investigate the effect of D-carnitine and tetradecylglycidic acid (TDGA) on palmitic acid absorption in the rat intestine.
    • To determine if D-carnitine or TDGA, a CPT inhibitor, impacts the intestinal uptake of palmitic acid.

    Main Methods:

    • Adult male rats underwent ligation of a proximal intestinal segment.
    • The segment was filled with 14C-palmitic acid, alone or with D-carnitine or TDGA.
    • Radioactivity was measured in lumenal contents, intestinal wall, and plasma after 30 minutes.

    Related Experiment Videos

    Main Results:

    • D-carnitine significantly decreased palmitic acid absorption, indicated by lower gut wall and plasma radioactivity and higher lumenal radioactivity.
    • L-carnitine alone had no effect on plasma radioactivity.
    • TDGA also demonstrated inhibition of intestinal palmitic acid absorption.

    Conclusions:

    • D-carnitine inhibits intestinal absorption of palmitic acid.
    • Tetradecylglycidic acid (TDGA), a carnitine palmitoyltransferase inhibitor, also reduces palmitic acid absorption.
    • These findings suggest carnitine palmitoyltransferase plays a role in intestinal fatty acid absorption.
