Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs01:25

Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs

407
Asthma is a chronic respiratory condition for which new therapeutic avenues, including anti-inflammatory drugs like mast cell stabilizers and anti-IgE treatments, continue to be developed.
Mast cell stabilizers, such as cromolyn (also known as sodium cromoglycate) and nedocromil (Tilade), are effective drugs in asthma management. These stabilizers hinder histamine release by skillfully obstructing the activation of mast cells and other cellular entities. Notably, they navigate this task without...

Immunoglobulin-like Cell Adhesion Molecules01:31

Immunoglobulin-like Cell Adhesion Molecules

3.4K
Immunoglobulin-like cell adhesion molecules or Ig-CAMs are a versatile group of cell surface glycoproteins belonging to the immunoglobulin protein superfamily. Ig-CAMs possess the characteristic immunoglobulin protein domains and other domains such as the fibronectin type III domain. The Ig domains are glycosylated to varying degrees in different Ig-CAMs.
Ig-CAMs exhibit either homophilic binding (to other Ig-CAMs) or heterophilic binding (to other ligands such as integrins). While most Ig-CAMs...

Antibody Structure

61.0K
Overview
Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding.
The Y-Shaped Structure of Antibodies Consists of Four Polypeptide Chains
Antibodies consist of four polypeptide chains: two identical heavy...

GPCRs Regulate Adenylyl Cylase Activity01:09

GPCRs Regulate Adenylyl Cylase Activity

5.8K
Some GPCRs transmit signals through adenylyl cyclase (AC), a transmembrane enzyme. AC helps synthesize second messenger cyclic adenosine monophosphate (cAMP). AC catalyzes cyclization reaction and converts ATP to cAMP by releasing a pyrophosphate. The pyrophosphate is further hydrolyzed to phosphate by the enzyme pyrophosphatase, which drives cAMP synthesis to completion. However, cAMP is rapidly degraded to 5′ AMP by the enzymes phosphodiesterase (PDE), preventing overstimulation of...

Transcytosis of IgG01:15

Transcytosis of IgG

2.8K
Transcytosis is the process in which molecules are internalized by endocytosis, transported across the cell, and released through exocytosis from the opposite end of the cell. Molecules such as insulin, immunoglobulins, and certain nutrients are transferred through the recycling endosomes by recycling and transcytosis.
IgG molecules from a mother undergo transcytosis starting around 13 weeks of gestation. The amount of IgG transferred and entering the fetal blood circulation increases with...

Selectins01:25

Selectins

3.4K
Cell adhesion is  an essential aspect of multicellularity. While stable cell interactions usually occur between cells of the same type, transient cell interactions occur between cells of different tissue types, such as between neutrophils and endothelial cells. Selectins are one class of cell adhesion molecules (CAMs) that bind carbohydrate ligands to form transient cell adhesion. They are rod-like proteins with a long extracellular part of variable length ending with the lectin domain,...

Summary

Nematode galectins inhibit mast cell degranulation, a key factor in gastrointestinal parasite survival. This study reveals how these parasite galectins reduce IgE activity, aiding nematode establishment and survival.

Area of Science:

  • Immunology
  • Parasitology
  • Molecular Biology

Background:

  • Mast cell degranulation is crucial for the survival of gastrointestinal nematodes like *Teladorsagia circumcincta*.
  • Host galectins (galectin-3 and -9) modulate mast cell degranulation by binding IgE.
  • Nematodes produce galectins, suggesting a potential role in immune evasion.

Purpose of the Study:

  • To investigate if nematode-derived galectins interfere with host galectins and inhibit mast cell degranulation.
  • To understand the immunomodulatory mechanisms employed by nematodes for establishment and survival.

Main Methods:

  • Enzyme-linked immunosorbent assay (ELISA) was used to quantify IgE activity.
  • A mast cell line was utilized to assess IgE binding and mediator release.
  • Levels of LCT-4, Beta-hexosaminidase, and MMP-9 were measured.

Main Results:

  • Nematode galectin significantly reduced total IgE activity.
  • Nematode galectin decreased the binding of sheep IgE to mast cells.
  • The release of LCT-4 and Beta-hexosaminidase was reduced, but MMP-9 release was unaffected.

Conclusions:

  • Nematode galectins play a role in modulating host mast cell degranulation.
  • This interaction represents a potential mechanism for nematodes to enhance their survival and establishment within the host.
  • Targeting this nematode immunomodulatory strategy could offer new approaches for parasite control.
