Insights

Increased TIGIT+PD-1+CXCR5-CD4+T cells are linked to rheumatoid arthritis (RA) pathogenesis. These cells correlate with autoantibodies and disease activity, suggesting a role in RA development.

Area of Science:

  • Immunology
  • Rheumatology
  • Cell Biology

Background:

  • Increased programmed cell death protein 1 (PD-1)+C-X-C chemokine receptor type 5 (CXCR5)- CD4+T cells are characteristic of rheumatoid arthritis (RA).
  • T cell immunoreceptor with immunoglobulin and ITIM domain (TIGIT) is a co-inhibitory receptor on T cells, but its role on PD-1+CXCR5-CD4+T cells in RA is unclear.

Purpose of the Study:

  • To investigate the expression patterns of TIGIT on PD-1+CXCR5-CD4+T cells in patients with RA.
  • To explore the association between TIGIT expression on these cells and clinical parameters of RA.

Main Methods:

  • Recruitment of patients with RA and healthy controls.
  • Flow cytometry to examine TIGIT expression on PD-1+CXCR5-CD4+T cells.
  • Correlation analysis between TIGIT+PD-1+CXCR5-CD4+T cell parameters and clinical/laboratory data, including autoantibodies and inflammatory markers.
  • Univariate and multivariate regression analyses to identify RA risk factors.

Main Results:

  • Percentages of TIGIT+CXCR5-CD4+T, PD-1+CXCR5-CD4+T, TIGIT+PD-1+CXCR5-CD4+T, and TIGIT-PD-1+CXCR5-CD4+T cells were significantly increased in RA patients.
  • Mean fluorescence intensity (MFI) of PD-1 and TIGIT on TIGIT+PD-1+CXCR5-CD4+T cells were also elevated in RA patients.
  • Parameters of TIGIT+PD-1+CXCR5-CD4+T cells correlated with autoantibodies (anti-CCP, RF) and inflammatory markers (ESR, CRP, various cell counts/ratios).
  • MFI of PD-1 in TIGIT+PD-1+CXCR5-CD4+T cells associated with disease activity scores (DAS28, VAS).
  • Multivariate analysis identified the percentage and MFI of PD-1 in TIGIT+PD-1+CXCR5-CD4+T cells as risk factors for RA.

Conclusions:

  • Elevated TIGIT+PD-1+CXCR5-CD4+T cells are significantly associated with autoantibody production and disease activity in RA.
  • These findings suggest a potential role for TIGIT+PD-1+CXCR5-CD4+T cells in the pathogenesis of rheumatoid arthritis.

T Cell Types and Functions01:24

T Cell Types and Functions

When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.

The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

T Cell Activation and Clonal Selection

T cells are integral to our adaptive immune system, recognizing and effectively responding to foreign antigens. T cell activation and clonal selection are pivotal in orchestrating this immune response. This article elucidates these mechanisms, detailing the roles of cluster of differentiation (CD) markers, major histocompatibility complex (MHC) molecules, costimulatory signals, and the process of clonal selection.

Drugs for Treatment of Crohn's Disease in IBD Using Biologic Agents: Anti-TNF01:24

Drugs for Treatment of Crohn's Disease in IBD Using Biologic Agents: Anti-TNF

Tumor Necrosis Factor (TNF), a proinflammatory cytokine, contributes significantly to the inflammation seen in Crohn's disease. It exists as soluble TNF and membrane-bound TNF, with actions mediated through TNF receptors (TNFR). TNFR activation leads to the release of proinflammatory cytokines, T-cell activation, collagen production, and leukocyte migration, all contributing to inflammation in Crohn's disease. Anti-TNF monoclonal antibodies, namely infliximab (Remicade), adalimumab...
