Genetic variants in the GARP gene are associated with house dust mite-sensitized persistent allergic rhinitis (PER) in Han Chinese. Specifically, SNP rs79525962 increases PER risk, particularly in females and those aged 16 and older.
Area of Science:
- Genetics
- Immunology
- Allergy Research
Background:
- Genetic variants in GARP (LRRC32) are linked to asthma and eczema.
- The association of GARP gene variants with allergic rhinitis is not well understood.
- This study focuses on house dust mite (HDM)-sensitized persistent allergic rhinitis (PER).
Purpose of the Study:
- To investigate the association between single nucleotide polymorphisms (SNPs) in the GARP gene and HDM-sensitized PER.
- To analyze GARP SNPs in a Han Chinese population.
Main Methods:
- A hospital-based case-control study involving 534 HDM-sensitized PER patients and 451 healthy controls.
- Identification of six SNPs within the GARP gene.
- Measurement of serum total and specific IgE levels.
- Prediction of RNA secondary structure and minimum free energy using RNAfold.
Main Results:
- SNP rs79525962 showed a significant association with the risk of HDM-sensitized PER.
- Individuals with CT+TT genotypes of rs79525962 had a higher risk (OR=1.393).
- The G-C haplotype of rs1320646-rs3781699 was associated with a lower risk (OR=0.819).
- rs79525962 increased PER risk in individuals aged ≥16 years and in females.
- GARP gene secondary structure was altered by different genotypes of rs79525962 and rs3781699.
Conclusions:
- SNP rs79525962 in the GARP gene represents a risk locus for HDM-sensitized PER in the Han Chinese population.
- Genetic variations in GARP may influence the development of allergic rhinitis.
