    Summary

    Sulpiride effectively increases prolactin levels even at low oral doses. Chronic sulpiride treatment, however, significantly reduces this prolactin-releasing effect over time.

    Background:

    • Prolactin is a hormone regulated by various factors, including dopamine.
    • Sulpiride is a dopamine antagonist known to affect prolactin secretion.

    Purpose of the Study:

    • To investigate the dose-response relationship of sulpiride on prolactin levels in healthy volunteers.
    • To assess the impact of circadian rhythms and chronic administration on sulpiride-induced prolactin release.

    Main Methods:

    • Healthy volunteers received oral doses of sulpiride (1 mg-50 mg) or placebo.
    • Prolactin levels were measured to determine acute and chronic responses.
    • Circadian variations in response were also evaluated.

    Main Results:

    • A 3 mg dose of sulpiride elicited prolactin levels comparable to 10 mg and 50 mg doses.
    • No significant differences in prolactin response were observed across different times of day (circadian effects).
    • Chronic sulpiride administration led to an attenuation of prolactin release, reaching approximately 50% of acute peak levels.

    Conclusions:

    • Sulpiride demonstrates potent prolactin-releasing activity even at low therapeutic doses.
    • The prolactin-releasing effect of sulpiride is diminished with chronic administration.
