Macrophage-capping protein (CAPG) promotes diffuse large B-cell lymphoma (DLBCL) cell proliferation and invasion by activating the PI3K/AKT pathway. CAPG is a potential therapeutic target for DLBCL treatment.
Area of Science:
- Oncology
- Molecular Biology
- Cancer Research
Background:
- Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous cancer.
- Macrophage-capping protein (CAPG) is implicated in various cancers.
- The role of CAPG in DLBCL remains largely unelucidated.
Purpose of the Study:
- To investigate the mechanism of CAPG in DLBCL.
- To determine CAPG's regulatory role and downstream effects in DLBCL.
Main Methods:
- Bioinformatic analysis for CAPG and target gene expression prediction.
- Quantitative real-time PCR (qRT-PCR), Western blot, and immunohistochemistry (IHC) for expression analysis.
- Flow cytometry for cell cycle and apoptosis, MTT assay for cell survival, and ChIP assay for gene binding.
Main Results:
- Elevated CAPG expression observed in DLBCL cells and tissues.
- CAPG enhances DLBCL cell proliferation and invasion while inhibiting apoptosis.
- CAPG activates the PI3K/AKT signaling pathway and accelerates DLBCL progression.
- CEBPA was identified as a regulator of CAPG expression.
Conclusions:
- CAPG promotes DLBCL proliferation and invasion via the PI3K/AKT pathway.
- CAPG represents a potential therapeutic target for DLBCL.

Tumor Progression
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
