MicroRNAs

3.1K
MicroRNA (miRNA) are short, regulatory RNA transcribed from introns (non-coding regions of a gene) or intergenic regions (stretches of DNA present between genes). Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself, forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA...

Area of Science:

  • Oncology
  • Molecular Biology
  • Biomarker Discovery

Background:

  • Gastric cancer lacks stable and validated biomarkers.
  • MicroRNAs (miRNAs) show promise as potential biomarkers.
  • Identifying specific miRNAs in gastric adenocarcinoma is crucial.

Purpose of the Study:

  • To identify and validate gastric cancer-specific miRNAs.
  • To determine alterations in miRNA levels in gastric adenocarcinoma tissues.
  • To use paired normal and tumor samples for validation.

Main Methods:

  • Studied miRNA expression in 20 paired gastric adenocarcinoma and normal stomach tissues.
  • Utilized quantitative reverse transcription polymerase chain reaction (RT-qPCR) for eight miRNAs.
  • Analyzed miRNA expression profiling in 40 tissue samples.

Main Results:

  • Six miRNAs (miR-375-3p, hsamiR-129-5p, miR-196a-5p, miR-376c-3p, miR-34c-5p, miR-767-5p) were significantly underexpressed in tumors.
  • miR-129-3p and miR34c-5p levels correlated with lymph node metastasis.
  • miR-376c-3p levels were negatively associated with smoking.

Conclusions:

  • Identified specific miRNAs as potential biomarkers for gastric cancer.
  • Alterations in miRNA expression can aid in developing diagnostic tools.
  • Lower miRNA expression variability in cancerous tissues compared to normal tissues was observed.
