Randomized Experiments

7.1K
The randomization process involves assigning study participants randomly to experimental or control groups based on their probability of being equally assigned. Randomization is meant to eliminate selection bias and balance known and unknown confounding factors so that the control group is similar to the treatment group as much as possible. A computer program and a random number generator can be used to assign participants to groups in a way that minimizes bias.

Regression Toward the Mean01:52

Regression Toward the Mean

6.4K
Regression toward the mean (“RTM”) is a phenomenon in which extremely high or low values—for example, and individual’s blood pressure at a particular moment—appear closer to a group’s average upon remeasuring. Although this statistical peculiarity is the result of random error and chance, it has been problematic across various medical, scientific, financial and psychological applications. In particular, RTM, if not taken into account, can interfere when...

Area of Science:

  • Genetics
  • Epidemiology
  • Causal Inference

Context:

  • Periodontitis is a complex inflammatory disease with largely observational evidence for associated factors.
  • Traditional observational studies are limited by confounding biases and reverse causality.
  • Mendelian randomization offers a robust approach to infer causality.

Purpose:

  • To review Mendelian randomization (MR) methodology.
  • To discuss the research progress of MR in the context of periodontitis.
  • To stimulate new research directions in periodontal disease etiology.

Summary:

  • Mendelian randomization (MR) leverages genetic variants as instrumental variables to infer causal relationships.
  • MR effectively mitigates confounding and reverse causality inherent in observational research.
  • This review highlights the growing application and advancements of MR in periodontitis research.

Impact:

  • Provides a framework for establishing causal links between various factors and periodontitis.
  • Enhances the reliability of etiological findings in periodontal research.
  • Opens avenues for developing targeted prevention and treatment strategies for periodontitis.
