Sp4 transcription factor regulates the PTTG1IP gene, which is involved in pituitary, thyroid, and breast cancer development. This study identified Sp4 binding sites and confirmed its role in PTTG-interacting protein gene expression.
Area of Science:
- Molecular Biology
- Oncology
- Gene Regulation
Background:
- Pituitary tumor-transforming gene 1 protein (PTTG)-interacting protein (PBF) is encoded by the proto-oncogene PTTG1IP.
- PBF interacts with PTTG and is implicated in pituitary, thyroid, and breast cancer etiology.
- The transcriptional regulation of the PTTG1IP gene is currently undefined.
Purpose of the Study:
- To investigate the transcriptional regulation of the human PTTG1IP gene.
- To identify the promoter region responsible for PTTG1IP gene activity.
- To determine the role of transcription factors in PTTG1IP gene expression.
Main Methods:
- Cloning and characterization of the 5' flanking region of the human PTTG1IP gene.
- Luciferase assay to identify the minimal promoter region (-212 to +7 bp).
- Electrophoretic mobility shift assay (EMSA) to detect transcription factor binding.
- Overexpression studies in HeLa cells to assess the impact of Sp4 on gene transcription.
Main Results:
- A 467-bp fragment of the 5' flanking region of the PTTG1IP gene was cloned.
- The region between -212 and +7 bp was identified as essential for PTTG1IP promoter activity.
- EMSA revealed Sp4 response elements within the PTTG1IP promoter.
- Overexpression of Sp4 significantly increased PTTG1IP gene transcription and expression in HeLa cells.
Conclusions:
- Sp4 acts as a transcriptional regulator of the PTTG1IP gene.
- Sp4 binds to specific response elements in the PTTG1IP promoter.
- These findings elucidate a key mechanism in the regulation of PTTG1IP, a gene involved in tumorigenesis.
