Background:

The kinome, the complete set of protein kinases, plays crucial roles in cellular signaling pathways in all organisms.
Understanding plant kinomes is essential for agricultural advancements and crop improvement.

Purpose of the Study:

To explore the functional conservation between human and plant kinases.
To identify novel chemical probes for dissecting plant kinase functions, particularly in root development.

Main Methods:

Cloning and expression of 129 rice (Oryza sativa) protein kinases in Escherichia coli.
Purification and screening of 40 rice kinases against 627 diverse human kinase inhibitors using differential scanning fluorimetry (DSF).
Testing of active compounds for effects on primary root development in Arabidopsis and identifying target plant kinases.

14 out of 37 active compounds significantly reduced primary root length in Arabidopsis.

Conclusions:

Human kinase inhibitor libraries are effective tools for identifying inhibitors of plant kinases.
The identified compounds can be used to investigate the roles of plant kinases, including those involved in root development.
Functional conservation exists between human and plant kinases, enabling cross-species application of chemical probes.
