Area of Science:

  • Neuroscience
  • Systems Neuroscience
  • Cognitive Neuroscience

Background:

  • Widespread coordination of spatially distant cortical areas forms functional networks crucial for cognition.
  • A common pattern involves co-activation of frontal and posterior cortical regions.
  • The precise neural circuit mechanisms for this inter-areal cortical coordination remain largely unknown.

Purpose of the Study:

  • To investigate the role of the claustrum in mediating frontoposterior cortical network coordination.
  • To elucidate the neural circuit mechanisms underlying widespread cortical communication.

Main Methods:

  • Utilized a discovery-based functional magnetic resonance imaging (fMRI) approach in a mouse model.
  • Examined functional connectivity between cortical regions and the subcortical claustrum.
  • Investigated synaptic connectivity within cortico-claustro-cortical circuits.

Main Results:

  • Identified significant functional connectivity between frontal and posterior cortical regions with the claustrum.
  • Observed distinct cortico-claustro-cortical circuits demonstrating trans-claustral synaptic connectivity.
  • Found that frontal cortices project to posterior sensory and sensory association cortices via the claustrum.

Conclusions:

  • The claustrum plays a key role in supporting frontoposterior cortical network architecture.
  • Discrete cortical pathways through the claustrum are positioned to facilitate cognitive control functions.
  • These findings contribute to understanding major extended cortico-subcortico-cortical systems in the mammalian brain.
