Area of Science:

  • Molecular Diagnostics
  • Virology
  • Point-of-Care Testing

Background:

  • Accurate and rapid detection of SARS-CoV-2 is crucial for controlling COVID-19 spread.
  • Current diagnostic methods may lack affordability, scalability, or ease of use for point-of-care applications.
  • There is a need for accessible diagnostic technologies for early infection detection.

Purpose of the Study:

  • To develop a rapid, efficient, and self-directed molecular diagnostic (SdMDx) system for SARS-CoV-2 detection.
  • To enable accurate and affordable point-of-care diagnosis of COVID-19.
  • To create a user-friendly system for self-operation and self-diagnosis.

Main Methods:

  • Developed a self-directed molecular diagnostic (SdMDx) system integrating sample preparation (virus enrichment/extraction) and detection.
  • Utilized dimethyl suberimidate dihydrochloride and functionalized diatomaceous earth for sample preparation.
  • Employed loop-mediated isothermal amplification-lateral flow assay (LAMP-LFA) for sensitive detection of the SARS-CoV-2 S gene.

Main Results:

  • The SdMDx system detected SARS-CoV-2 within 47 minutes without specialized instruments.
  • Achieved a limit of detection as low as 0.05 PFU in culture fluid.
  • Clinical validation on 38 nasopharyngeal specimens showed 94.4% sensitivity and 100% specificity for the S gene.

Conclusions:

  • The SdMDx system provides a rapid, accurate, and simple method for SARS-CoV-2 detection.
  • This technology is suitable for point-of-care testing in various settings, including low-resource areas and home use.
  • The SdMDx system offers a sensitive alternative to antigen and antibody assays, minimizing contamination risks.
