Summary

Methotrexate treatment caused significant damage to the jejunum's autonomic nerves. Both adrenergic and cholinergic nerve profiles showed degeneration, with prolonged use leading to substantial nerve loss and damage to nerve cells.

Area of Science:

  • Gastroenterology
  • Neuroscience
  • Pharmacology

Background:

  • Autonomic nerves regulate gastrointestinal function.
  • Methotrexate is a chemotherapy agent with known side effects.

Purpose of the Study:

  • To investigate the impact of Methotrexate on the autonomic nerve supply of the jejunum.
  • To quantify nerve degeneration following Methotrexate administration.

Main Methods:

  • Histochemical analysis of jejunum tissue.
  • Daily injection of 10 mg/kg Methotrexate for 3 weeks (5 days/week).

Main Results:

  • Partial degeneration of adrenergic and cholinergic nerves observed after 1 week.
  • Approximately 30% adrenergic and 50% cholinergic nerve profile loss after 3 weeks.
  • Early degenerative changes noted in intestinal plexus cholinergic nerve cells.

Conclusions:

  • Methotrexate induces significant dose-dependent damage to jejunal autonomic innervation.
  • Damage affects both adrenergic and cholinergic systems, impacting nerve cells within the intestinal plexuses.
  • Findings highlight potential mechanisms for Methotrexate-induced gastrointestinal dysfunction.
