DropLab is a new point-of-care testing (POCT) platform that uses magnetic digital microfluidics (MDM) to perform enzyme-linked immunosorbent assays (ELISAs) in droplets. This automated system offers sensitive and quantitative results for rapid clinical decisions.
Area of Science:
- Biomedical Engineering
- Microfluidics
- Immunodiagnostics
Background:
- Point-of-care testing (POCT) demands rapid, accurate diagnostic results near patients.
- Current POCT immunodiagnostics, like lateral flow assays (LFAs), lack sensitivity and quantification.
- Traditional enzyme-linked immunosorbent assays (ELISAs) offer sensitivity but require complex liquid handling unsuitable for POCT.
Purpose of the Study:
- To develop an automated, sample-in-answer-out POCT immunodiagnostic platform.
- To leverage magnetic digital microfluidic (MDM) technology for enhanced ELISA performance.
- To create a user-friendly device for sensitive and quantitative biomarker detection.
Main Methods:
- Developed DropLab, an automated POCT platform utilizing MDM technology.
- Implemented microbead-based ELISA within digitally controlled droplets.
- Designed a thermoformed disposable chip for parallel assay processing (4 ELISAs).
Main Results:
- DropLab achieved sensitive and quantitative results comparable to conventional microwell plate ELISAs.
- The platform demonstrated a limit of detection (LOD) matching traditional ELISA.
- Validated performance by measuring two protein targets and one antibody target.
Conclusions:
- DropLab successfully integrates MDM technology for automated, high-throughput POCT immunodiagnostics.
- The platform overcomes limitations of existing POCT assays, offering improved sensitivity and quantification.
- DropLab represents a significant advancement towards real-world medical applications of MDM technology.
