This study introduces a smart dressing that simultaneously promotes blood vessel growth (angiogenesis) and nerve regeneration for improved wound healing. The innovative design integrates multiple therapeutic agents for enhanced, in-situ tissue repair.
Area of Science:
- Biomaterials Science
- Regenerative Medicine
- Nanotechnology
Background:
- Wound repair and skin appendage regeneration are hindered by inadequate angiogenesis and neural regeneration.
- Current therapeutic strategies often address angiogenesis and neural regeneration separately, limiting effectiveness.
- A combined approach is needed for simultaneous proangiogenic and neuro-regenerative effects in wound healing.
Purpose of the Study:
- To investigate the efficacy of an all-in-one smart dressing (ASD) for simultaneous angiogenesis and neural regeneration.
- To develop a multi-functional hydrogel-based dressing integrating angiogenic and neuro-regenerative therapeutic agents.
- To demonstrate the potential of a novel nanomaterial-drug combination for enhanced wound repair.
Main Methods:
- Development of a light crosslinked hydrogel incorporating zeolite-imidazolate framework 67 with anchored vanadium oxide (VO2@ZIF-67).
- Incorporation of CXCL12, ligustroflavone, and ginsenoside Rg1 for sustained release of therapeutic agents.
- Evaluation of the ASD's ability to stimulate angiogenesis via pH-triggered Co2+ release and neural regeneration through sustained drug delivery.
Main Results:
- The ASD demonstrated on-demand release of Co2+ from VO2@ZIF-67, stimulating angiogenesis.
- Sustained release of CXCL12, ligustroflavone, and ginsenoside Rg1 promoted mesenchymal stem cell recruitment, inhibited senescence, and induced neural differentiation.
- The smart dressing facilitated rapid angiogenesis and nerve regeneration within 17 days.
Conclusions:
- The developed all-in-one smart dressing effectively integrates nanomaterials and multiple drugs for simultaneous angiogenesis and neural regeneration.
- This study provides a proof-of-concept for a novel therapeutic system to overcome limitations in wound repair and regeneration.
- The ASD offers a promising strategy for advanced wound management by addressing critical aspects of tissue repair concurrently.
