Area of Science:

  • Hematology
  • Clinical Chemistry
  • Medical Technology

Background:

  • Light transmission aggregometry (LTA) is the gold standard for assessing platelet function.
  • Current LTA methods are labor-intensive and manual, limiting standardization.
  • Automation presents an opportunity to improve LTA standardization and accessibility.

Purpose of the Study:

  • To evaluate the performance characteristics of the automated Thrombomate XRA (TXRA) instrument.
  • To compare TXRA's performance against a manual instrument (PAP-8) for platelet function testing.
  • To assess TXRA's correlation with both autologous and "virtual" platelet-poor plasma (VPPP).

Main Methods:

  • Parallel testing of leftover blood samples using both manual PAP-8 and automated TXRA.
  • Evaluation of precision and method comparison for maximum aggregation (MA%) values.
  • Comparison of TXRA results against autologous platelet-poor plasma (PPP) and AI-based VPPP.

Main Results:

  • TXRA demonstrated good precision for MA% (1.4-4.6%) across all reagents.
  • Normal ranges on TXRA were similar to the manual method, with slightly higher values observed.
  • Excellent correlation was found between TXRA, manual methods, and VPPP, with similar reaction signatures.

Conclusions:

  • The automated TXRA instrument provides reproducible LTA results that correlate well with established manual methods.
  • TXRA simplifies LTA by requiring only platelet-rich plasma, eliminating the need for autologous PPP.
  • TXRA represents a significant advancement for standardizing and broadening the use of platelet function analysis.
