Background:

  • Raynaud's phenomenon is a condition causing reduced blood flow to extremities.
  • Dihydropyridine derivatives are commonly used for cardiovascular conditions.

Purpose of the Study:

  • To evaluate the efficacy and tolerability of nisoldipine in treating primary Raynaud's phenomenon.
  • To compare nisoldipine's effects with placebo in a controlled trial.

Main Methods:

  • A placebo-controlled, double-blind, cross-over trial was conducted.
  • 36 patients with primary Raynaud's phenomenon received nisoldipine (5 mg and 10 mg daily) or placebo for one month each.

Main Results:

  • Nisoldipine did not lead to subjective symptom improvement or changes in finger blood flow.
  • No significant alterations in platelet aggregability or red cell deformability were observed.
  • Adverse effect incidence was comparable to nifedipine, indicating sufficient plasma concentrations.

  • Nisoldipine is ineffective for treating primary Raynaud's phenomenon at doses up to 10 mg/day.
  • Unlike nifedipine, nisoldipine does not appear to be a viable therapeutic option for this condition.
