Praxelis clematidea, an invasive plant, yielded new compounds with anti-inflammatory potential. Certain compounds significantly reduced nitric oxide (NO) production and NF-κB nuclear translocation, suggesting therapeutic applications.
Area of Science:
- Phytochemistry
- Pharmacology
- Immunology
Background:
- Praxelis clematidea is a noxious invasive plant in southern China, causing ecological and economic damage.
- Natural products research aims to identify bioactive compounds from plants for therapeutic development.
Purpose of the Study:
- To isolate and characterize chemical constituents from Praxelis clematidea.
- To evaluate the anti-inflammatory potential of isolated compounds by assessing their effects on nitric oxide (NO) production and NF-κB nuclear translocation in macrophages.
Main Methods:
- Phytochemical analysis involving separation and purification of compounds from the whole plant using spectroscopic methods.
- In vitro assays using lipopolysaccharide (LPS)-triggered RAW 264.7 macrophages to evaluate NO production, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2) expression, and NF-κB nuclear translocation.
Main Results:
- Six new compounds (four phenolics and two phenylpropanoids) and seventeen known compounds were isolated and identified.
- Compounds 2, 7, and 8 demonstrated significant inhibition of NO production.
- Compounds 2, 7, and 8 also suppressed iNOS and COX-2 expression and inhibited NF-κB nuclear translocation.
Conclusions:
- Praxelis clematidea contains bioactive compounds with notable anti-inflammatory properties.
- The isolated compounds, particularly 2, 7, and 8, show promise for the development of treatments for inflammation-related diseases.
