Background:

  • Propionic acid (PPA) administration in rats induces autism-like behaviors and neuroinflammation.
  • PPA exposure leads to elevated pro-inflammatory cytokines (TNF-α, IL-17, IL-2), Nerve Growth Factor (NGF), and oxidative stress markers (MDA, lactate).
  • Histopathological changes, including altered neuronal counts and GFAP immunoreactivity, are observed in PPA-treated rats.

Purpose of the Study:

  • To investigate the potential of Modafinil in mitigating PPA-induced neuroinflammation, behavioral deficits, and histopathological alterations.
  • To assess the impact of Modafinil on key biochemical markers associated with autism and neuroinflammation.

Main Methods:

  • Thirty Wistar rats were divided into three groups: control, PPA + saline, and PPA + Modafinil.
  • Autism was induced in two groups using daily intraperitoneal injections of PPA (250 mg/kg/day) for 5 days.
  • Modafinil (30 mg/kg/day) or saline was administered orally to the respective groups.
  • Behavioral tests, biochemical analyses (cytokines, NGF, MDA, lactate), immunohistochemistry (neurons, GFAP), and Magnetic Resonance Spectroscopy (MRS) were employed.

Main Results:

  • PPA administration induced significant autism-like behaviors, increased levels of TNF-α, NGF, IL-17, IL-2, MDA, and lactate, and altered neuronal counts and GFAP immunoreactivity.
  • MRS revealed elevated lactate and decreased creatinine levels in PPA-treated rats.
  • Modafinil treatment significantly reversed the behavioral, neuroinflammatory, and histopathological changes induced by PPA.

Conclusions:

  • Modafinil demonstrates a significant ameliorating effect on PPA-induced autism-like behaviors and neuroinflammation in a rat model.
  • The findings suggest Modafinil's therapeutic potential for conditions associated with PPA-induced neuroinflammation.
  • Further research is warranted to explore Modafinil's mechanisms of action in this context.
