Area of Science:

  • Virology
  • Immunology
  • Tropical Medicine

Background:

  • Dengue virus (DENV) is a significant global health threat, causing over 400 million infections yearly, with severe forms like DHF and DSS.
  • No specific antiviral treatments exist, making vaccine development crucial for controlling this mosquito-borne flavivirus.

Purpose of the Study:

  • To review the current landscape of dengue vaccine development.
  • To highlight the progress and efficacy of recently licensed vaccines, Dengvaxia (CYD-TDV) and Denvax (TAK003).

Main Methods:

  • Review of clinical trial data for CYD-TDV and TAK003.
  • Analysis of vaccine efficacy in different age groups and prior infection statuses.
  • Discussion of ongoing vaccine research and development.

Main Results:

  • CYD-TDV demonstrates high efficacy in children aged 9+ with prior DENV exposure, but carries risks for younger seronegative children.
  • TAK003 showed high efficacy against DENV2 (97.7%) and DENV1 (73.7%) in Phase 3 trials in children aged 4-16.
  • Other vaccines like TV003 and TV005 are in earlier stages of development.

Conclusions:

  • CYD-TDV and TAK003 represent significant advancements in dengue vaccine technology.
  • These novel vaccines offer potential for controlling dengue, a major neglected tropical disease (NTD).
  • Continued research is vital to develop broadly protective and safe dengue vaccines for all populations.
