Summary

Genomic selection of Gir embryos using in vitro fertilization (IVF) is feasible. High-density imputation improved genomic estimated breeding value (GEBV) accuracy for milk yield, enabling early selection for improved traits.

Area of Science:

  • Animal Genetics
  • Reproductive Technologies
  • Bioinformatics

Background:

  • Genomic selection (GS) is crucial for accelerating genetic gain in livestock.
  • In vitro fertilization (IVF) offers opportunities for early genetic assessment of embryos.
  • Establishing a reliable platform for genomic selection in Gir embryos is needed.

Purpose of the Study:

  • To develop a genomic selection platform for IVF Gir embryos.
  • To assess the accuracy of genomic estimated breeding values (GEBV) derived from embryo biopsies.
  • To evaluate the impact of imputation methods on GEBV accuracy.

Main Methods:

  • Multiple displacement amplification (MDA) for whole-genome amplification of embryo biopsy DNA.
  • Genotyping of embryo and liveborn samples using Z-Chip and Bovine HD panels.
  • Imputation using large reference populations (73,110 lactating cows, 147,131 pedigrees).
  • Calculation and comparison of GEBV for 305-day milk yield (305MY).

Main Results:

  • Biopsied embryos showed comparable pregnancy and loss rates to controls.
  • Bovine HD imputation achieved higher accuracy (0.82) for embryo GEBV compared to Z-Chip (0.55) or no imputation (0.62).
  • High correlation (0.87) between embryo and calf GEBV estimates was observed with Bovine HD imputation.
  • Embryo sample call rate did not impact GEBV accuracy post-imputation.

Conclusions:

  • Genomic selection of Gir embryos via IVF is a viable strategy for genetic improvement.
  • High-density imputation significantly enhances the accuracy of early genomic predictions.
  • This platform facilitates early identification of superior genetics, impacting milk yield in the Gir breed.
