Adenosine's role in regulating basal coronary arteriolar tone.

H Gewirtz, R A Olsson, D L Brautigan

    Summary

Adenosine plays a limited role in regulating coronary arteriolar tone under basal conditions. Adenosine deaminase infusion showed minimal impact on regional blood flow, suggesting limited influence of adenosine in normal coronary circulation.

    Area of Science:

    • Cardiovascular Physiology
    • Pharmacology

    Background:

    • Adenosine is a nucleoside that influences vascular tone.
    • Its role in regulating basal coronary arteriolar tone requires further elucidation.

    Purpose of the Study:

    • To investigate the role of adenosine in regulating coronary arteriolar tone under basal conditions in a normal coronary circulation model.

    Main Methods:

    • Hemodynamic measurements and microsphere-based flow analysis were performed in pigs.
    • Adenosine deaminase (ADA) was infused into the left anterior descending (LAD) coronary artery to degrade adenosine.
    • Interstitial fluid penetration and adenosine concentration reduction by ADA were assessed in swine.

    Main Results:

    • Infusion of ADA did not significantly alter transmural flow in the LAD zone.

    Related Experiment Videos

  • ADA infusion led to a significant increase in the LAD:circumflex flow resistance ratio.
  • Regional myocardial oxygen consumption remained unchanged.
  • ADA effectively penetrated interstitial fluid and reduced basal adenosine concentrations.
  • Conclusions:

    • Adenosine appears to play a limited role in regulating coronary arteriolar tone under basal conditions in a healthy coronary circulation.
    • The observed modest changes in flow resistance suggest other factors are more dominant in basal tone regulation.
