Background:

  • The 2022 global outbreak of monkeypox virus (MPXV) highlighted the urgent need for effective treatments beyond limited smallpox vaccines and antivirals.
  • Orthopoxviruses, including MPXV, pose significant public health threats, necessitating the exploration of novel therapeutic strategies.

Purpose of the Study:

  • To identify novel antiviral compounds with inhibitory activity against orthopoxviruses, specifically vaccinia virus (VACV) and MPXV.
  • To evaluate compounds previously shown to inhibit RNA viruses for their potential efficacy against DNA viruses like orthopoxviruses.

Main Methods:

  • Utilized recombinant vaccinia virus (rVACV) reporter systems (fluorescence and luciferase) to screen 19 compounds from the ReFRAME and NPC libraries for antiorthopoxvirus activity.
  • Confirmed the inhibitory activity of promising compounds against both rVACV and MPXV in vitro.

  • Seven compounds from the ReFRAME library and six from the NPC library demonstrated inhibitory activity against rVACV.
