    Summary

    Ciliary abnormalities are common in ependymoma, with many structurally abnormal cilia found in unusual locations within tumor cells. These findings suggest a potential oncogenetic effect on cilia formation in ependymomas.

    Background:

    • Ependymal cells normally have cilia with a 9+2 microtubule structure.
    • Cilia are typically sparse or absent in ependymomas, and their abnormalities are poorly documented.

    Purpose of the Study:

    • To investigate the ultrastructure and incidence of ciliary abnormalities in ependymal cells of a fourth ventricular ependymoma.
    • To quantitatively analyze ciliary location and structural defects.

    Main Methods:

    • Detailed ultrastructural examination of cilia from a pediatric ependymoma.
    • Quantitative analysis of cilia location (extracellular microrosettes, intracytoplasmic lumina, cytoplasm) and structural abnormalities.

    Main Results:

    • 30% of 267 studied cilia exhibited structural abnormalities.
    • Aberrantly located cilia (intracytoplasmic, cytoplasmic) showed higher abnormality rates.
    • Common abnormalities included abnormal axial microtubules, compound cilia, swollen cilia, and dynein arm defects.

    Conclusions:

    • Ciliary abnormality is frequent in ependymoma.
    • Abnormalities may stem from oncogenetic effects impacting ciliogenesis in ciliated epithelium.
