In human women, oogenesis produces one mature egg cell or ovum for every precursor cell that enters meiosis. Background:

Purpose of the Study:

  • To localize the maturation-promoting factor (MPF) within starfish oocytes.
  • To identify the subcellular components associated with MPF activity.

Main Methods:

  • Stratification of ooplasm from maturing starfish oocytes using density-graded Ficoll-seawater centrifugation.
  • Injection of stratified ooplasm into immature oocytes to assay for MPF activity.
  • Electron microscopic examination of oocyte strata.

Main Results:

  • MPF activity was predominantly found in the hyaline layer stratum.
  • The hyaline layer was rich in Golgi complexes, lucent vesicles, and multi-vesicular bodies.
  • Other strata (oil cap, yellow layer) contained different inclusions and showed less MPF activity.

Conclusions:

  • The hyaline layer of starfish oocytes is the primary location of MPF.
  • Specific organelles within the hyaline layer, such as Golgi complexes and vesicles, are likely associated with MPF activity.
  • This study provides insights into the subcellular localization of key factors regulating oocyte maturation.
