Ribosome Profiling02:24

Ribosome Profiling

3.6K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.

Area of Science:

  • Microbiology
  • Plant Pathology
  • Biotechnology

Background:

  • Trichoderma longibrachiatum SMF2 is a known biocontrol agent.
  • Understanding its mechanism is crucial for agricultural applications.

Purpose of the Study:

  • To identify and analyze effector proteins secreted by T. longibrachiatum SMF2.
  • To elucidate the role of these proteins in biocontrol and plant interaction.

Main Methods:

  • Bioinformatics analysis of secreted proteins.
  • Transcriptome sequencing to identify upregulated proteins.
  • Quantitative PCR validation of specific effector candidates.

Main Results:

  • Identified 478 secretory proteins from T. longibrachiatum.
  • Found 272 proteins upregulated after plant treatment.
  • 36 secretory proteins showed homology to known effectors from pathogens.

Conclusions:

  • Secretory proteins from T. longibrachiatum SMF2 likely function as effectors.
  • These effectors may promote fungal growth, colonization, or induce plant immunity.
