lncRNA - Long Non-coding RNAs

8.7K
In humans, more than 80% of the genome gets transcribed. However, only around 2% of the genome codes for proteins. The remaining part produces non-coding RNAs which includes ribosomal RNAs, transfer RNAs, telomerase RNAs, and regulatory RNAs, among other types. A large number of regulatory non-coding RNAs have been classified into two groups depending upon their length – small non-coding RNAs, such as microRNA, which are less than 200 nucleotides in length, and long non-coding RNA...

Abnormal Proliferation02:23

Abnormal Proliferation

4.6K
Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the...

Summary

B-cell leukemia/lymphoma 11A (BCL11A) is upregulated in non-small cell lung cancer (NSCLC). Its nuclear expression correlates with proliferation markers and may drive tumor progression, while cytoplasmic expression shows inverse relationships.

Area of Science:

  • Oncology
  • Molecular Biology
  • Biomarker Research

Background:

  • B-cell leukemia/lymphoma 11A (BCL11A) is a potential biomarker for non-small cell lung cancer (NSCLC).
  • The precise role of BCL11A in NSCLC development requires further elucidation.
  • Understanding BCL11A expression patterns is crucial for identifying therapeutic targets.

Purpose of the Study:

  • To investigate BCL11A expression at mRNA and protein levels in NSCLC versus non-malignant lung tissue (NMLT).
  • To correlate BCL11A expression with clinicopathological factors and proliferation markers (Ki-67, Slug, Snail, Twist).
  • To determine the differential localization (nuclear vs. cytoplasmic) of BCL11A in NSCLC subtypes.

Main Methods:

  • Immunohistochemistry (IHC) and immunofluorescence (IF) for protein expression analysis.
  • Real-time PCR for mRNA expression quantification.
  • Analysis of 259 NSCLC cases and 116 NMLT samples, including cell line studies.

Main Results:

  • BCL11A protein expression was significantly elevated in NSCLC compared to NMLT.
  • Nuclear BCL11A expression was observed in squamous cell carcinoma (SCC), while cytoplasmic expression was noted in adenocarcinoma (AC).
  • Nuclear BCL11A decreased with higher malignancy grades and positively correlated with Ki-67 and Slug/Twist; cytoplasmic expression showed opposite correlations.

Conclusions:

  • BCL11A expression is altered in NSCLC, with distinct localization in different subtypes.
  • Nuclear BCL11A may promote NSCLC progression by influencing cell proliferation and phenotype.
  • BCL11A warrants further investigation as a potential therapeutic target in NSCLC.
