    Summary

    An acid-stable trypsin inhibitor was found in bile from patients with malignant tumors and gallstones. This bile trypsin inhibitor shares antigenicity with urinary trypsin inhibitor and may have clinical significance.

    Background:

    • Acid-stable trypsin inhibitor identified in bile of patients with malignant tumors (biliary tract/pancreas head carcinoma) and gallstones.

    Purpose of the Study:

    • To characterize the bile trypsin inhibitor found in malignant tumor patients.
    • To investigate its biochemical properties and potential clinical significance.

    Main Methods:

    • Partial purification using DEAE cellulose ion exchange chromatography.
    • Molecular weight determination by gel filtration.
    • Isoelectric focusing and analytical polyacrylamide gel electrophoresis for characterization.
    • Enzyme inhibition assays (trypsin, chymotrypsin, plasmin, urokinase).

    • Two molecular forms identified: a major form (MW ~86,000) and a minor form (MW ~31,000).

    • The larger form showed cross-reactivity with anti-urinary trypsin inhibitor serum.

    • Bile contains an acid-stable trypsin inhibitor antigenically related to urinary trypsin inhibitor.
    • The inhibitor exists in at least two molecular forms with distinct properties.
    • Further investigation into the clinical significance of this bile trypsin inhibitor is warranted.
